On the activity of praziquantel against larvae of Clonorchis sinensis experimentally infected in rats

Abstract
The present study was conducted to observe a chemoprophylactic activity of praziquantel (Biltricide(D) against early larval stage of Clonorchis sinensis.
The rats experimentally infected with about 50 metacercariae of C. sinensis per animal were treated with doses of 3 x 100 mg/kg x 2 days, 1 x 300 mg/kg x 1 day and 1 x 600 mg/kg x 1 day of praziquantel at 4 and 24 hours before or after infection respectively, and the curative effects of the drug were compared with those at 5 weeks (adult worms) after infection.
Sixty-three rats divided into 15 groups including a control rat in each group were autopsied at 4 weeks (29 days) after the infection in order to evaluate the curative activity of the drug based on recovery rate of the worms.
The results obtained are as follows:
1. In the groups treated at 24 hours before infection, a single dose of 300 mg/kg and 600 mg/kg of praziquantel showed evident prophylactic effects and only a few worms were recovered in each one out of each 3 rats(worm recovery rates of 1.8 % and 17.2% respectively). However, in dividing doses of 3 x 100 mg/kg x 2 days of praziquantel, no marked difference of the worm recovery rates was shown in comparison with that of control rat.
2. In the groups treated at 4 hours before infection with the doses of 3 x 100 mg/kg x 2 days and 1 x 300 mg/kg x 1 day of praziquantel, a few worms were recovered in each 2 out of 3 rats and the worm recovery rates were shown as 1.8 to 14. 0 per cent. But a group treated with a single dose of 600 mg/kg of praziquantel showed complete cure.
3. In the groups treated at 4 and 24 hours after infection, all rats treated with all kinds of dosages of praziquantel were cured completely. On the other hand, in the groups treated at 5 weeks after infection with a single dose of 300 mg/kg and 600 mg/kg of praziquantel, only one worm (1.8% worm recovery rate) from each one out of 3 rats was recovered. In the group treated with dividing doses of 3 x 100 mg/kg x 2 days, however, all rats were cured completely.
From the above results, praziquantel showed a high degree of activity against larvae as well as adult worms of C. sinensis. However, a single dose of medication showed more prophylactic effect than that of dividing doses at 24 hours before infection.
